چکیده

Optical Coherence Tomography (OCT) and Terahertz pulsed imaging (TPI) are two powerful techniques allowing high quality cross-sectional images from within scattering media to be obtained non-destructively. In this paper, we report experimental results of using OCT and TPI for quantitatively characterizing pharmaceutical tablet coatings in the thickness range of 10—140 μm. We found that the spectral OCT system developed in-house has an axial resolution of 0.9 μm, and is capable of quantifying very thin coatings in the range of 10—60 μm. The upper limit of 60 μm within the tablet coating and core is owing to the strong scattering of OCT light, which has relatively short wavelengths in the range of 0.5— 1.0 μm. On the other hand, TPI utilizes terahertz radiation which has substantially longer wavelengths in the range of hundreds microns, thus is less prone to the scattering problem. Consequently TPI has been demonstrated to be able to quantify thicker coatings in the range of 40—140 μm and beyond. We concluded that OCT and TPI are two complementary analytical techniques for non-destructive and quantitative characterizing pharmaceutical tablet coatings.

Quantifying Pharmaceutical Film Coating with Optical Coherence Tomography and Terahertz Pulsed Imaging: An Evaluation

Spectral domain optical coherence tomography (OCT) has recently attracted a lot of interest in the pharmaceutical industry as a fast and non-destructive modality for quantification of thin film coatings that cannot easily be resolved with other techniques. Because of the relative infancy of this technique, much of the research to date has focused on developing the in-line measurement technique ...

Imaging pharmaceutical tablets with optical coherence tomography.

Optical coherence tomography (OCT) is a recently developed optical technique that produces depth profiles of three-dimensional objects. It is a nondestructive interferometric method responding to refractive index variation in the sample under study and can reach a penetration depth of a few millimetres. OCT employs near-infrared (NIR) light and therefore provides a link between NIR spectroscopy...

Terahertz Technology: A Boon to Tablet Analysis

The terahertz gap has a frequency ranges from approximately 0.3 THz to approximately 10 THz in the electromagnetic spectrum which is in between microwave and infrared. The terahertz radiations are invisible to naked eye. In comparison with x-ray they are intrinsically safe, non-destructive and non-invasive. Terahertz spectroscopy enables 3D imaging of structures and materials, and the measureme...

A comparison of quality control methods for active coating processes.

Terahertz pulsed imaging (TPI) is a recent and nondestructive technique to quantify coating thickness of pharmaceutical tablet film coatings. In this study, TPI is used for the first time to quantify the progress of an active coating process. The dosage form consisted of a push-pull osmotic system comprising a two-layer tablet core with a functional film coating and a laser drilled hole. On top...

Non-destructive evaluation of polymer coating structures on pharmaceutical pellets using full-field optical coherence tomography.

Full-field optical coherence tomography (FF-OCT) using a conventional light-emitting diode and a complementary metal-oxide semiconductor camera has been developed for characterising coatings on small pellet samples. A set of en-face images covering an area of 700 × 700 μm(2) was taken over a depth range of 166 μm. The three-dimensional structural information, such as the coating thickness and u...
